Yes, religion and politics, after England and Spain became enemies, due to the religious reformation in England. We should not forget the attempt to marry the future Charles I to Infanta Maria. He even travelled to Madrid to meet the Infanta. A lot has been written about that trip.
After the Reformation I don't think any member of the Spanish royal family married a Protestant (unlike the Stuart kings who all, but one, took Catholic wives). To fanatical Catholics like the Habsburgs, such unions were unthinkable. The attempt by the Prince of Wales (the future Charles I) to woo the Infanta was famously farcical and never seriously contemplated by the Spanish court.
Ena of Battenberg was the first bride from a Protestant country to marry a Spanish monarch and, of course, she had to change her religion before any wedding could take place.
