You would fly into Philly airport. Depending on how adventurous you are, you could take the R1 regional rail from the airport into downtown and then a taxi to the Alexander Inn ... or you can take a taxi from the airport.
I stayed at the Alexander Inn when I went a couple of years ago, and I thought it was really nice and very reasonable.
You definitely will NOT need a car. There is much to do right in the gayborhood, and even if you wanted to go see the museums or whatever, they are very walk-able. In fact, folks from Philly, myself included, tend not to have cars. Parking is a pain and gas is expensive.
In terms of what to do... there is a lot to do in the area. If you want to get the full gayborhood experience, I would go to one of the local restaurants for dinner, including:
Mixto's - GREAT cuban food
El Vez - Stephen Starr mexican restaurant
Raw - possibly the best sushi I have ever had!
Then I would head to 'bump' for drinks (gay bar with FABULOUS white chocolate martinis); then - if you have never been to a lesbian bar - I would go to Sisters. There is a bar on the main level, pool tables downstairs, and dance floor upstairs. If you have been to a lesbian bar, I recommend skipping Sisters (I am not really a fan) and going to Woody's (a gay bar around the corner) - it has a bar downstairs and dance floor upstairs - it tends to really be packed and fun on Saturday nights. On friday nights, they have country line dancing. It is a gay bar - so lots of gay men - but still a mixture of folks there.
If you have time during the day, I recommend checking out Giovanni's room - an lgbt bookstore right down the street from the Alexander Inn.
Philly is a great city - you will have a blast!!
